These counts and lists are worked out from the library itself each time the page loads. A score rates how much weight a study can bear — its design, journal, evidence and recency — not whether its result is good news. How we score a study. For a plain-English guide to this subject, see Treatments.

The 20 highest-scoring studies

  1. Efficacy and safety of passive immunotherapies targeting amyloid beta in Alzheimer's disease: A systematic review and meta-analysis PLoS medicine · 2025-03-31 · · Peer-reviewed

  5. TRAILBLAZER-ALZ 4: A phase 3 trial comparing donanemab with aducanumab on amyloid plaque clearance in early, symptomatic Alzheimer's disease Alzheimer's & dementia : the journal of the Alzheimer's Association · 2025-05-01 · · Peer-reviewed
  6. Efficacy of anti-amyloid-ß monoclonal antibody therapy in early Alzheimer's disease: a systematic review and meta-analysis Neurological sciences : official journal of the Italian Neurological Society and of the Italian Society of Clinical Neurophysiology · 2023-11-17 · · Peer-reviewed
  7. Two phase 3 trials of bapineuzumab in mild-to-moderate Alzheimer's disease The New England journal of medicine · 2014-01-01 · · Peer-reviewed

In two large phase 3 trials, the anti-amyloid antibody bapineuzumab given to people with mild-to-moderate Alzheimer's disease produced no significant difference from placebo on the main measures of cognition and daily function. This means the drug did not work as hoped in these patients, and it is not a treatment for Alzheimer's disease.

  8. Effects of monoclonal antibodies against amyloid-β on clinical and biomarker outcomes and adverse event risks: A systematic review and meta-analysis of phase III RCTs in Alzheimer's disease Ageing research reviews · 2021-04-05 · · Peer-reviewed

This analysis of donanemab trials found that amyloid-related imaging abnormalities (ARIA) are an adverse event associated with donanemab treatment that requires safety monitoring. Several baseline factors were linked to higher or lower risk of ARIA-E.

  15. Amyloid-Related Imaging Abnormalities in 2 Phase 3 Studies Evaluating Aducanumab in Patients With Early Alzheimer Disease JAMA neurology · 2022-01-01 · · Peer-reviewed

This report describes amyloid-related imaging abnormalities in two phase 3 studies of aducanumab in people with early Alzheimer disease. The provided text does not include information on whether aducanumab affected cognitive or functional outcomes.

  16. Randomized Trial of Verubecestat for Prodromal Alzheimer's Disease The New England journal of medicine · 2019-04-01 · · Peer-reviewed

This trial tested verubecestat, a BACE-1 inhibitor that blocks production of amyloid-beta, in patients with prodromal Alzheimer's disease. The drug did not prevent clinical progression in an earlier trial involving patients with mild-to-moderate dementia due to Alzheimer's disease.
