These counts and lists are worked out from the library itself each time the page loads. A score rates how much weight a study can bear — its design, journal, evidence and recency — not whether its result is good news.

  1. Integrating a multimodal lifestyle intervention with medical food in prodromal Alzheimer's disease: the MIND-ADmini randomized controlled trial Alzheimer's research & therapy · 2024-05-30 · · Peer-reviewed
  2. A randomized, double-blind, phase 2b proof-of-concept clinical trial in early Alzheimer's disease with lecanemab, an anti-Aβ protofibril antibody Alzheimer's research & therapy · 2021-04-17 · · Peer-reviewed

This phase 2b trial tested lecanemab, an antibody targeting soluble aggregated amyloid beta, in people with early Alzheimer's disease, mild cognitive impairment due to Alzheimer's disease, and mild Alzheimer's disease dementia, using a Bayesian design to find the effective dose. It is a proof-of-concept trial and does not establish that lecanemab cures or prevents Alzheimer's disease.

  3. evoke and evoke+: design of two large-scale, double-blind, placebo-controlled, phase 3 studies evaluating efficacy, safety, and tolerability of semaglutide in early-stage symptomatic Alzheimer's disease Alzheimer's research & therapy · 2025-01-08 · · Peer-reviewed

This paper sets out the design of evoke and evoke+, two large randomized, double-blind, placebo-controlled phase 3 trials of once-daily oral semaglutide versus placebo in people with early-stage symptomatic Alzheimer's disease. The trials enrolled participants between May 18, 2021, and September 8, 2023, to test efficacy, safety and tolerability. It is a design paper: it does not report whether semaglutide helped, so it should not be read as evidence that the drug works.

  4. Lecanemab in patients with early Alzheimer's disease: detailed results on biomarker, cognitive, and clinical effects from the randomized and open-label extension of the phase 2 proof-of-concept study Alzheimer's research & therapy · 2022-12-21 · · Peer-reviewed
  5. Central autonomic network dysfunction and plasma Alzheimer's disease biomarkers in older adults Alzheimer's research & therapy · 2024-06-08 · · Peer-reviewed
  6. A multidomain lifestyle intervention to maintain optimal cognitive functioning in Dutch older adults-study design and baseline characteristics of the FINGER-NL randomized controlled trial Alzheimer's research & therapy · 2024-06-13 · · Peer-reviewed
  7. Inflammatory proteins associated with Alzheimer's disease reduced by a GLP1 receptor agonist: a post hoc analysis of the EXSCEL randomized placebo controlled trial Alzheimer's research & therapy · 2024-10-02 · · Peer-reviewed
  8. Updated safety results from phase 3 lecanemab study in early Alzheimer's disease Alzheimer's research & therapy · 2024-05-10 · · Peer-reviewed

This report gives updated safety results from a phase 3 lecanemab study in early Alzheimer's disease, covering 1795 participants in the core study and 1612 who received at least one dose. It is about safety data and does not report on whether the drug cures or prevents the disease.

  9. A phase III randomized trial of gantenerumab in prodromal Alzheimer's disease Alzheimer's research & therapy · 2017-12-08 · · Peer-reviewed

In this 2-year phase III trial (SCarlet RoAD), 797 people with prodromal Alzheimer's disease were randomized to monthly injections of gantenerumab 105 mg, 225 mg or placebo. The trial was stopped early for futility, with no difference from placebo on the main measure of decline (CDR-SB) or on secondary clinical measures. Brain-scan changes called ARIA, mostly without symptoms, became more common with higher dose and in APOE ε4 carriers; the authors' suggestion that higher doses might be needed comes from exploratory analyses only.

  10. Effects of functional tasks exercise on cognitive functions of older adults with mild cognitive impairment: a randomized controlled pilot trial Alzheimer's research & therapy · 2019-12-04 · · Peer-reviewed
  11. 27-Hydroxycholesterol, cognition, and brain imaging markers in the FINGER randomized controlled trial Alzheimer's research & therapy · 2021-03-06 · · Peer-reviewed
  12. Efficacy and safety of GV1001 in patients with moderate-to-severe Alzheimer's disease already receiving donepezil: a phase 2 randomized, double-blind, placebo-controlled, multicenter clinical trial Alzheimer's research & therapy · 2021-03-26 · · Peer-reviewed
  13. A phase II randomized trial of sodium oligomannate in Alzheimer's dementia Alzheimer's research & therapy · 2020-09-14 · · Peer-reviewed
  14. Brain volumes and cortical thickness on MRI in the Finnish Geriatric Intervention Study to Prevent Cognitive Impairment and Disability (FINGER) Alzheimer's research & therapy · 2019-06-04 · · Peer-reviewed
  15. Masitinib for mild-to-moderate Alzheimer's disease: results from a randomized, placebo-controlled, phase 3, clinical trial Alzheimer's research & therapy · 2023-02-28 · · Peer-reviewed

This phase 3 trial tested masitinib, an oral drug targeting immune cells in the brain, as an add-on to existing dementia medications in people with mild-to-moderate Alzheimer's disease. It was a randomized, placebo-controlled study, so its results are needed to judge whether this drug affects the disease course.

  16. Combining plasma phospho-tau and accessible measures to evaluate progression to Alzheimer's dementia in mild cognitive impairment patients Alzheimer's research & therapy · 2022-03-29 · · Peer-reviewed
  17. Efficacy of the Ubiquitous Spaced Retrieval-based Memory Advancement and Rehabilitation Training (USMART) program among patients with mild cognitive impairment: a randomized controlled crossover trial Alzheimer's research & therapy · 2017-06-06 · · Peer-reviewed
  18. A 36-week multicenter, randomized, double-blind, placebo-controlled, parallel-group, phase 3 clinical trial of sodium oligomannate for mild-to-moderate Alzheimer's dementia Alzheimer's research & therapy · 2021-03-17 · · Peer-reviewed

In this 36-week trial of 818 people with mild-to-moderate Alzheimer's dementia, those taking sodium oligomannate (GV-971) showed a significant difference in cognitive scores favoring the drug compared with placebo. The study reports this as an improvement in cognition, but it did not show that the drug cures or prevents Alzheimer's disease.

  19. Effects of intensive lifestyle changes on the progression of mild cognitive impairment or early dementia due to Alzheimer's disease: a randomized, controlled clinical trial Alzheimer's research & therapy · 2024-06-07 · · Peer-reviewed

In this randomized controlled trial of 51 people with mild cognitive impairment or early Alzheimer's dementia, an intensive multidomain lifestyle intervention was compared with usual care over 20 weeks. The summary states that comprehensive lifestyle changes may significantly improve cognition and function, but this is a single small trial and does not establish a cure or prevention.

  20. A phase 2 double-blind placebo-controlled 24-week treatment clinical study of the p38 alpha kinase inhibitor neflamapimod in mild Alzheimer's disease Alzheimer's research & therapy · 2021-05-27 · · Peer-reviewed

In this 24-week phase 2 trial in mild Alzheimer's disease, the drug neflamapimod did not improve the main or secondary clinical measures compared with placebo. It did significantly lower spinal-fluid levels of total tau and p-tau 181, markers of nerve damage and tau disease. Combined with signals in people with the highest drug levels, the authors argue a longer, higher-dose study is warranted, so any clinical benefit remains unproven.
