Alzheimer’s disease (AD) is the most widespread neurodegenerative disorder and a leading cause of dementia seen in clinical practice.
Why it matters
This review covers established and emerging Alzheimer's drugs, including cholinesterase inhibitors, memantine, several monoclonal antibodies, nutritional supplements, and agents targeting tau and amyloid pathways. It is a summary of existing research rather than a new trial, so it does not by itself show that any treatment cures or prevents the disease.
-
A 77-year-old woman with mild cognitive impairment was referred to 18F-florbetapir positron emission tomography (PET) to assess the indication of a disease-modifying drug against Alzheimer disease.
Why it matters
This case report describes a failed 18F-florbetapir PET scan caused by an air-vent filter trapping the radioactivity, which was resolved by using a different filter. It is a technical imaging issue and has no bearing on treating, curing, or preventing Alzheimer's disease.
-
Alzheimer disease (AD) is a major health issue of global concern whereby there are minimal treatment options.
Why it matters
This review proposes that the dietary carotenoid lutein could act as a 'molecular sponge' neutralizing toxic aldehydes such as 4-HNE and acrolein that injure neurons. This is a hypothesis about a possible mechanism, not evidence that lutein treats, cures, or prevents Alzheimer's disease.

As a result of multiple phosphorylation, Tau undergoes early conformational changes involving the bending of the intact amino (N)-terminus over the third repeated domain, which in turn was evidenced by positive immunolabeling with the monoclonal antibody Alz-50 [2].
Why it matters
This laboratory work describes molecular events in the tau protein, including phosphorylation-driven conformational changes and ubiquitination in neurofibrillary tangles, that may correlate better with neuropathological progression of the disease. It also reports altered tubulin cytoskeleton structure in neuroblastoma cells after transfection, which is early cell-based research and not a treatment finding.
