Relationship Between Plasma Neurofilament Light Chain, Gut Microbiota, and Dementia: A Cross-Sectional Study
Naoki Saji, Kenta Murotani, Naoyuki Sato, Tsuyoshi Tsuduki, Takayoshi Hisada, Mitsuru Shinohara, Taiki Sugimoto, Shumpei Niida et al.. Journal of Alzheimer's disease : JAD, 2022
Topic: Gut, metabolism and diet
We analyzed 128 participants (women: 59%, mean age: 74 years). Participants with high (above the median) plasma NfL concentrations tended to be older, women, and hypertensive and have a history of stroke, chronic kidney disease, and dementia. Plasma NfL was also associated with cerebral small vessel disease. However, plasma NfL levels were not significantly correlated with gut microbial metabolites. Multivariable analyses revealed that a higher plasma NfL concentration was independently associated with the presence of dementia (odds ratio: 9.94, 95% confidence interval: 2.75-48.2, p < 0.001). High plasma NfL concentration was independently associated with the presence of dementia as previously reported. However, plasma NfL levels were not significantly correlated with gut microbial metabolites in this preliminary study.

Key findings
- We analyzed 128 participants (women: 59%, mean age: 74 years).
- Participants with high (above the median) plasma NfL concentrations tended to be older, women, and hypertensive and have a history of stroke, chronic kidney disease, and dementia.
- Plasma NfL was also associated with cerebral small vessel disease.
- However, plasma NfL levels were not significantly correlated with gut microbial metabolites.
- Multivariable analyses revealed that a higher plasma NfL concentration was independently associated with the presence of dementia (odds ratio: 9.94, 95% confidence interval: 2.75-48.2, p < 0.001).

How this study scored 66/100
Published in a peer-reviewed journal, and it is a cohort study following people over time. Scored zero for effect reported and randomised / blinded because the record does not state it. We have no data on journal's citation record, times cited, citations per year, impact for its field and year, cited by studies here, still being cited, senior author's track record, institution's track record, multi-centre collaboration and free to read, so those were left out: it earned 37 of the 56 points we could judge it on, which is 66 out of 100.
