  1. A tetravalent TREM2 agonistic antibody reduced amyloid pathology in a mouse model of Alzheimer's disease

✓ Peer-reviewed alzhub usefulness score 52 out of 100. Science translational medicine 2022-09-07 TREM2 and immune genetics

Triggering receptor expressed on myeloid cells 2 (TREM2) plays crucial roles in Alzheimer's disease (AD) by regulating microglia migration toward, and phagocytosis of oligomeric amyloid-β (oAβ) and amyloid plaques.

    Why it matters

A TREM2 agonist antibody, made stronger by engineering, increased microglial phagocytosis and reduced amyloid pathology in a mouse model of Alzheimer's disease. This is animal research on a potential drug approach and has not been shown to work in people.

  2. CSF phosphorylated tau as an indicator of subsequent tau accumulation

✓ Peer-reviewed alzhub usefulness score 52 out of 100. Neurobiology of aging 2022-05-26 Biomarkers

We evaluated the relationship between baseline CSF p-tau181 and the rate of tau PET change in the temporal meta-ROI and entorhinal cortex (ERC) and how it varied by amyloid level (CSF Aβ42 or amyloid PET) among 143 individuals from the Mayo Clinic Study of Aging and Mayo Alzheimer Disease Research Center.

    Why it matters

Among 143 people, higher CSF p-tau181 and amyloid markers were linked to faster tau PET accumulation, with the entorhinal cortex changing more in non-demented participants. This is an observational imaging study and does not test any treatment, cure, or prevention.

  3. Disease-linked TDP-43 hyperphosphorylation suppresses TDP-43 condensation and aggregation

✓ Peer-reviewed alzhub usefulness score 52 out of 100. The EMBO journal 2022-02-03 Brain tissue and pathology

Post-translational modifications (PTMs) have emerged as key modulators of protein phase separation and have been linked to protein aggregation in neurodegenerative disorders.

    Why it matters

This laboratory work found that Casein kinase 1delta-mediated TDP-43 hyperphosphorylation or C-terminal phosphomimetic mutations reduced TDP-43 phase separation and aggregation, making condensates more liquid-like and dynamic, contrary to the usual assumption. It is early mechanistic research on TDP-43 in ALS and frontotemporal dementia and does not address treating, curing or preventing Alzheimer's disease.

  4. Selective reduction of astrocyte apoE3 and apoE4 strongly reduces Aβ accumulation and plaque-related pathology in a mouse model of amyloidosis

This study reveals an important role of astrocytic apoE3 and apoE4 on the deposition and accumulation of Aβ plaques as well as on certain Aβ-associated downstream effects.

    Why it matters

In a mouse model of amyloidosis, selectively reducing astrocyte apoE3 and apoE4 strongly reduced amyloid-beta plaque deposition and produced less compact plaques. This is an animal study and reports no treatment, cure or prevention in people.

  5. Neuropathology and molecular diagnosis of Synucleinopathies

✓ Peer-reviewed alzhub usefulness score 52 out of 100. Molecular neurodegeneration 2021-12-18 Getting a diagnosis

Synucleinopathies are clinically and pathologically heterogeneous disorders characterized by pathologic aggregates of α-synuclein in neurons and glia, in the form of Lewy bodies, Lewy neurites, neuronal cytoplasmic inclusions, and glial cytoplasmic inclusions.

    Why it matters

This review sorts the α-synuclein diseases into two groups: Lewy body disease (Parkinson's disease, Parkinson's disease with dementia and dementia with Lewy bodies) and multiple system atrophy. It describes how lab seeding tests such as RT-QuIC and PMCA "offer exciting opportunities for molecular diagnosis using readily accessible peripheral tissue samples," but it reports no accuracy figures. It also says "there are currently no disease-modifying therapies for the synucleinopathies," and it does not address Alzheimer's disease directly.

✓ Peer-reviewed alzhub usefulness score 52 out of 100. Alzheimer's & dementia (Amsterdam, Netherlands) 2019-08-28 Digital health tools

This multimodal approach enhanced the system's performance and provided a clinically informative method for neuroimaging analysis.

    Why it matters

In 44 people with behavioural-variant frontotemporal dementia and 60 healthy people scanned at three centres with different MRI setups, a fully automated computer pipeline combining brain-structure and resting-brain-activity scans sorted people into the right group 91% of the time (it caught 83.7% of patients and correctly cleared 96.6% of healthy people). The study compared patients only with healthy people, not with Alzheimer's or other conditions that look similar, so it shows promise for a research tool rather than a ready clinical test. It does not test any treatment, cure or prevention.

  17. Small-Molecule Modulation of TDP-43 Recruitment to Stress Granules Prevents Persistent TDP-43 Accumulation in ALS/FTD

    Why it matters

In a mouse model of Alzheimer's disease, the Caspase-1 inhibitor VX-765 reversed memory impairment and hyperactivity, but memory deficits returned about a month after the drug was stopped. This is early animal research, so it does not yet show that the drug would work or be safe in people.

  22. C9orf72-mediated ALS and FTD: multiple pathways to disease

✓ Peer-reviewed alzhub usefulness score 52 out of 100. Nature reviews. Neurology 2018-09-01 Lewy body dementia

The discovery that repeat expansions in the C9orf72 gene are a frequent cause of amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) has revolutionized our understanding of these diseases.

    Why it matters

This review proposes that C9orf72-related ALS and FTD arise from a combination of loss and gain of function plus downstream cellular pathways. It is a summary of mechanisms, not a trial, and suggests no specific treatment.

  23. In Vivo Characterization and Quantification of Neurofibrillary Tau PET Radioligand 18F-MK-6240 in Humans from Alzheimer Disease Dementia to Young Controls

  25. Age-Dependent Effects of apoE Reduction Using Antisense Oligonucleotides in a Model of β-amyloidosis

    ✓ Peer-reviewed alzhub usefulness score 52 out of 100. Neuron 2017-12-01 Genetics

The apolipoprotein E (APOE) gene is the strongest genetic risk factor for late-onset Alzheimer disease.

    Why it matters

In APP/PS1-21 mice, reducing apoE with an antisense oligonucleotide before plaque deposition decreased amyloid pathology, while starting treatment at the onset of amyloid deposition increased plaque size and reduced plaque-associated neuritic dystrophy without changing overall plaque load. These are mouse results showing that timing of apoE reduction matters, and they do not establish a treatment for people.
