The Association Between Polygenic Risk for Alzheimer's, Objective Sleep Architecture, and Plasma Neurofilament Light Chain in Midlife
Asma Riffaqat, Sumera Kanwal, Zoha Zahra, Ali Hamza Mirza, Kiran Naved Khan, Shama Nosherwani, Maaz Ubair Shaikh. Journal of Health Wellness and Community Research, 2026-01-15
Topic: Amyloid, tau and blood-test diagnosis
PRS correlated positively with log-NFL (r = 0.31, p = 0.004) and inversely with N3 duration (r = −0.26, p = 0.017). The PRS × N3 interaction was negative (β = −0.31, p = 0.003), indicating a stronger PRS–NFL association at shorter N3 durations. The PRS × log-WASO interaction was not statistically significant (β = 0.14, p = 0.190). N3 duration modified the observed association between genetic susceptibility and plasma NFL. These cross-sectional findings do not establish causality or Alzheimer-specific pathology.

Key findings
- PRS correlated positively with log-NFL (r = 0.31, p = 0.004) and inversely with N3 duration (r = −0.26, p = 0.017).
- The PRS × N3 interaction was negative (β = −0.31, p = 0.003), indicating a stronger PRS–NFL association at shorter N3 durations.
- The PRS × log-WASO interaction was not statistically significant (β = 0.14, p = 0.190).
- N3 duration modified the observed association between genetic susceptibility and plasma NFL.
- These cross-sectional findings do not establish causality or Alzheimer-specific pathology.
Why it matters
In midlife, higher polygenic risk for Alzheimer's correlated with higher plasma neurofilament light chain, and this association was stronger at shorter N3 sleep duration. The authors state these cross-sectional findings do not establish causality or Alzheimer-specific pathology.

How this study scored 38/100
Published in a peer-reviewed journal, and it is a snapshot of people at one point in time. Scored zero for effect reported and randomised / blinded because the record does not state it. We have no data on senior author's track record, so that was left out: it earned 36 of the 96 points we could judge it on, which is 38 out of 100.
