Peripheral risk factors and their role in biomarker-based screening for dementia in the community
Yi-Han Hu, Osorio Meirelles, Eric J Shiroma, Claudia L Satizabal, Sudha Seshadri, Russell P Tracy, Vilmundur Gudnason, Lenore J Launer. Alzheimer's & dementia : the journal of the Alzheimer's Association, 2024-12
Topic: Plasma and spinal-fluid biomarkers
Besides age and estimated glomerular filtration rate (eGFR), significant associations were found between plasma biomarkers and clusters related to hemoglobin, red blood cell distribution, and inflammation. Incorporating these clusters into predictive models enhanced precision and sensitivity, though overall prediction improvement was modest (area under the precision-recall curve: GFAP 0.17 to 0.34, NfL 0.20 to 0.38). PRFs are significantly associated with dementia plasma biomarkers; Considering these factors may enhance the predictive accuracy of dementia biomarkers. HIGHLIGHTS: Machine learning identifies key peripheral factors influencing neurodegenerative biomarkers. Hemoglobin and red blood cell distribution cluster associates significantly with biomarker levels. Incorporating diverse peripheral factors modestly enhances incident dementia prediction accuracy in community settings.

How this study scored 55/100
Published in a leading specialty journal. Scored zero for study design, effect reported and randomised / blinded because the record does not state it. We have no data on journal's citation record, times cited, citations per year, impact for its field and year, cited by studies here, still being cited, senior author's track record, institution's track record, multi-centre collaboration and free to read, so those were left out: it earned 31 of the 56 points we could judge it on, which is 55 out of 100.
