Perivascular spaces are associated with tau pathophysiology and synaptic dysfunction in early Alzheimer's continuum
Natalia Vilor-Tejedor, Iacopo Ciampa, Grégory Operto, Carles Falcón, Marc Suárez-Calvet, Marta Crous-Bou, Mahnaz Shekari, Eider M Arenaza-Urquijo et al.. Alzheimer's research & therapy, 2021-08-05
Topic: Plasma and spinal-fluid biomarkers
The degree of PVS significantly increased with age in both, BG and CS regions independently of cardiovascular risk factors. Higher levels of p-tau, t-tau, and neurogranin were significantly associated with ePVS in the CS of Aβ positive individuals, after accounting for relevant confounders. No associations were detected in the BG neither in Aβ negative participants. Our results support that ePVS in the CS are specifically associated with tau pathophysiology, neurodegeneration, and synaptic dysfunction in asymptomatic stages of the Alzheimer's continuum.
Key findings
- The degree of PVS significantly increased with age in both, BG and CS regions independently of cardiovascular risk factors.
- Higher levels of p-tau, t-tau, and neurogranin were significantly associated with ePVS in the CS of Aβ positive individuals, after accounting for relevant confounders.
- No associations were detected in the BG neither in Aβ negative participants.
- Our results support that ePVS in the CS are specifically associated with tau pathophysiology, neurodegeneration, and synaptic dysfunction in asymptomatic stages of the Alzheimer's continuum.

How this study scored 50/100
Published in a peer-reviewed journal. Scored zero for study design, effect reported and randomised / blinded because the record does not state it. We have no data on journal's citation record, times cited, citations per year, impact for its field and year, cited by studies here, still being cited, senior author's track record, institution's track record, multi-centre collaboration and free to read, so those were left out: it earned 28 of the 56 points we could judge it on, which is 50 out of 100.
