Misfolded alpha synuclein co-occurrence with Alzheimer's disease proteinopathy
Erin M Jonaitis, Karen MacLeod, Jennifer Lamoureux, Beckie Jeffers, Rachel L Studer, John Middleton, Rachael E Wilson, Nathaniel A Chin et al.. medRxiv : the preprint server for health sciences, 2024-10-13
Topic: Plasma and spinal-fluid biomarkers
Syn positivity (synSAA+) co-occurred with T (in synSAA+ vs synSAA-, 36% vs 20% T+; p=0.011) and with cognitive impairment (10% vs 7% MCI; 10% vs 0% dementia; p=0.00050). synSAA+ participants' cognitive performance declined ∼40% faster than synSAA-for Digit Symbol, but not other tests. Findings support prevalent syn copathology in a mostly-unimpaired AD risk cohort. Future work will explore relationships with disease progression.

Key findings
- Syn positivity (synSAA+) co-occurred with T (in synSAA+ vs synSAA-, 36% vs 20% T+; p=0.011) and with cognitive impairment (10% vs 7% MCI; 10% vs 0% dementia; p=0.00050). synSAA+ participants' cognitive performance declined ∼40% faster than synSAA-for Digit Symbol, but not other tests.
- Findings support prevalent syn copathology in a mostly-unimpaired AD risk cohort.
- Future work will explore relationships with disease progression.

How this study scored 16/100
Preprint — not yet peer-reviewed. It scored 45 on the same rubric, but non-peer-reviewed sources are capped at 16, so it shows as 16. Scored zero for effect reported and randomised / blinded because the record does not state it. We have no data on times cited, citations per year, impact for its field and year, cited by studies here, still being cited, senior author's track record, institution's track record, multi-centre collaboration and free to read, so those were left out: it earned 27 of the 60 points we could judge it on, which is 45 out of 100.
| Component | Points | Why |
|---|---|---|
| Journal tier | 2 / 10 | Tier 4 — not a peer-reviewed journal (medRxiv : the preprint server for health sciences) |
| Journal's citation record | 0 / 4 | Not a journal article, so the journal's record does not count |
| Study design | 7 / 12 | A longitudinal study following the same people over time |
| Strength of evidence | 3 / 5 | Rated “moderate” in our summary |
| Peer-review validation | 4 / 10 | Preprint — not yet peer-reviewed. Hosted on a preprint server. |

| Points | 27 / 60 | Scored on 11 of 20 parts. We have no data for the rest, so they are left out: 27 of the 60 points we could judge it on is 45 out of 100. |
| Total | 45 / 100 | Capped to 16 because this is not a peer-reviewed article. |
