Ibuprofen effects on Alzheimer pathology and open field activity in APPsw transgenic mice
Lim GP, Yang F, Chu T, Gahtan E, Ubeda O, Beech W, Overmier JB, Hsiao-Ashec K et al.. Neurobiology of aging, 2001-11-01
Topic: Brain proteins, microglia and lab models
We previously showed the non-steroidal anti-inflammatory drug (NSAID) ibuprofen suppresses inflammation and amyloid in the APPsw (Tg2576) Tg2576 transgenic mouse. The mechanism for these effects and the impact on behavior are unknown. We now show ibuprofen's effects were not mediated by alterations in amyloid precursor protein (APP) expression or oxidative damage (carbonyls).
Key findings
- The mechanism for these effects and the impact on behavior are unknown.
- We now show ibuprofen's effects were not mediated by alterations in amyloid precursor protein (APP) expression or oxidative damage (carbonyls).
- Six months ibuprofen treatment in Tg+ females caused a decrease in open field behavior (p < 0.05), restoring values similar to Tg- mice.
- Reduced caspase activation per plaque provided further evidence for a neuroprotective action of ibuprofen.
Why it matters
This study found that six months of ibuprofen treatment in transgenic mice reduced open field behavior and showed reduced caspase activation per plaque, but the effects were not mediated by changes in APP expression or oxidative damage. It is animal research and does not show that ibuprofen treats or prevents Alzheimer's disease in people.

How this study scored 44/100
Published in a peer-reviewed journal. Scored zero for study design, sample size, effect reported and randomised / blinded because the record does not state it. We have no data on senior author's track record, so that was left out: it earned 42 of the 96 points we could judge it on, which is 44 out of 100.
