✓ Peer-reviewed alzhub usefulness score 35 out of 100. Tau biomarkers

Topic: Amyloid, tau and blood-test diagnosis

Soluble phosphorylated tau has become a key biomarker for Alzheimer's disease pathology, yet further mechanistic studies are needed beyond observational investigations to clarify the relationship between soluble p-tau species and intracellular tau pathology. Here, we utilized chimeric human cerebral organoids (chCOs) to generate a series of organoids in which the endogenous MAPT gene was CRISPR/Cas9 edited in different cell types to create phosphorylation-deficient mutants at the AD-associated sites. We found that the APPswe mutation increased tau phosphorylation in both neurons and astrocytes.

Key findings

Why it matters

Using engineered human cerebral organoids, this laboratory study found that astrocyte-specific phosphorylation-deficient mutations of tau reduced soluble p-tau181 and p-tau217 levels. This is early, cell-based mechanistic work that helps explain what p-tau biomarkers reflect, not a treatment or cure.

Total 35 of 100.34 of the 96 points we could judge = 35 of 100

Published in a peer-reviewed journal. Scored zero for study design, sample size, effect reported and randomised / blinded because the record does not state it. We have no data on senior author's track record, so that was left out: it earned 34 of the 96 points we could judge it on, which is 35 out of 100.
