A DAP12-mediated pathway regulates expression of CC chemokine receptor 7 and maturation of human dendritic cells
Bouchon A, Hernández-Munain C, Cella M, Colonna M.. The Journal of experimental medicine, 2001-10-01
Topic: Brain proteins, microglia and lab models
Gene targeting of the adaptor molecule DAP12 in mice caused abnormal distribution and impaired antigen presentation capacity of dendritic cells (DCs). However, the DAP12-associated receptors expressed on DCs and their functions have not been identified yet. Here we show that the triggering receptor expressed on myeloid cells-2 (TREM-2) is a cell surface receptor on human monocyte-derived DCs, which is associated with DAP12.

Key findings
- However, the DAP12-associated receptors expressed on DCs and their functions have not been identified yet.
- Here we show that the triggering receptor expressed on myeloid cells-2 (TREM-2) is a cell surface receptor on human monocyte-derived DCs, which is associated with DAP12.
- TREM-2/DAP12 promotes upregulation of CC chemokine receptor 7, partial DC maturation, and DC survival through activation of protein tyrosine kinases and extracellular signal-regulated kinase.
- In contrast to Toll-like receptor-mediated signaling, TREM2/DAP12 stimulation is independent of nuclear factor-kappaB and p38 stress-activated protein kinase.
Why it matters
In human immune cells grown from blood monocytes (dendritic cells), this 2001 lab study reports that TREM-2 is a surface receptor that works through the adaptor DAP12. Triggering it raised the receptor CCR7, produced "partial DC maturation" and helped the cells survive, using a signalling route the authors say differs from Toll-like receptors. It is basic immunology. It does not study the brain, microglia or people with Alzheimer's, and it reports no numbers.

How this study scored 52/100
Published in a peer-reviewed journal. Scored zero for study design, sample size, effect reported, studied in people and randomised / blinded because the record does not state it. We have no data on institution's track record, so that was left out: it earned 50 of the 97 points we could judge it on, which is 52 out of 100.
