Polymer-Engineered Aβ-Degrading Proteases via Photo-ATRP for Alzheimer's Disease Therapeutics
Andrey Tsoy, Sholpan Askarova, Bibifatima Kaupbayeva. Eurasian journal of applied biotechnology, 2025-11-14
Topic: Amyloid, tau and blood-test diagnosis
BSA-polymer conjugates displayed enhanced resistance to proteolytic degradation. Successful polymer growth confirmed by molecular weight shifts. These results validated the protein-compatibility and shielding effect of the polymer grafts under biologically relevant conditions. This study positions as the first application of photo-ATRP "grafting-from" polymerization to engineer Aβ-degrading enzymes. The polymer conjugation significantly improves protein stability and immune evasion while retaining catalytic potential. This platform presents a promising strategy for next-generation enzyme-based therapeutics in AD, addressing critical limitations of current antibody-based approaches. The integration of polymer chemistry and neurobiology opens new pathways for creating catalytically active, long-circulating, and biocompatible therapeutic enzymes. Key findings
- BSA-polymer conjugates displayed enhanced resistance to proteolytic degradation.
- Successful polymer growth confirmed by molecular weight shifts.
- These results validated the protein-compatibility and shielding effect of the polymer grafts under biologically relevant conditions.
- This study positions as the first application of photo-ATRP "grafting-from" polymerization to engineer Aβ-degrading enzymes.
- The polymer conjugation significantly improves protein stability and immune evasion while retaining catalytic potential.
Why it matters
This laboratory study used polymer chemistry to make amyloid-beta-degrading enzymes more stable and better able to avoid immune detection. It is early laboratory work and does not show any effect on Alzheimer's disease in people.

How this study scored 27/100
Published in a peer-reviewed journal. Scored zero for study design, sample size, effect reported, studied in people and randomised / blinded because the record does not state it. We have no data on institution's track record, so that was left out: it earned 26 of the 97 points we could judge it on, which is 27 out of 100.
