Higher CSF sTREM2 attenuates ApoE4-related risk for cognitive decline and neurodegeneration
Franzmeier N, Suárez-Calvet M, Frontzkowski L, Moore A, Hohman TJ, Morenas-Rodriguez E, Nuscher B, Shaw L et al.. Molecular neurodegeneration, 2020-10-08
Topic: Brain proteins, microglia and lab models
Background The Apolipoprotein E ε4 allele (i.e. ApoE4) is the strongest genetic risk factor for sporadic Alzheimer's disease (AD). Triggering receptor expressed on myeloid cells 2) is a microglial transmembrane protein brain that plays a central role in microglia activation in response to AD brain pathologies.
Key findings
- ApoE4) is the strongest genetic risk factor for sporadic Alzheimer's disease (AD).
- Triggering receptor expressed on myeloid cells 2) is a microglial transmembrane protein brain that plays a central role in microglia activation in response to AD brain pathologies.
- Whether higher TREM2-related microglia activity modulates the risk to develop clinical AD is an open question.
- Thus, the aim of the current study was to assess whether higher sTREM2 attenuates the effects of ApoE4-effects on future cognitive decline and neurodegeneration.
Why it matters
In a longitudinal study, higher CSF sTREM2 attenuated the link between ApoE4 and future cognitive decline and neurodegeneration. This is an observational finding and does not yet show a treatment, cure, or prevention for Alzheimer's disease.

How this study scored 62/100
Published in a peer-reviewed journal, and it is a longitudinal study following the same people over time. Scored zero for effect reported and randomised / blinded because the record does not state it.
