Alzheimer's disease genetic risk variants beyond APOE ε4 predict mortality
Jesse Mez, Jessica R Marden, Shubhabrata Mukherjee, Stefan Walter, Laura E Gibbons, Alden L Gross, Laura B Zahodne, Paola Gilsanz et al.. Alzheimer's & dementia (Amsterdam, Netherlands), 2017-08-14
Topic: Genetics and APOE
Higher GRS predicted mortality (hazard ratio = 1.05; 95% confidence interval: 1.00-1.10, P = .04). After adjusting for cognitive performance or restricting to participants with dementia, the relationship was attenuated and no longer significant. In case-control analysis, the GRS was associated with reduced longevity (odds ratio = 0.64; 95% confidence interval: 0.41-1.00, P = .05). Non-APOE LOAD susceptibility loci confer risk for mortality, likely through effects on dementia incidence.
Key findings
- Higher GRS predicted mortality (hazard ratio = 1.05; 95% confidence interval: 1.00-1.10, P = .04).
- After adjusting for cognitive performance or restricting to participants with dementia, the relationship was attenuated and no longer significant.
- In case-control analysis, the GRS was associated with reduced longevity (odds ratio = 0.64; 95% confidence interval: 0.41-1.00, P = .05).
- Non-APOE LOAD susceptibility loci confer risk for mortality, likely through effects on dementia incidence.

How this study scored 57/100
Published in a leading specialty journal, and it is a case-control study. Scored zero for sample size, effect reported, studied in people and randomised / blinded because the record does not state it. We have no data on journal's citation record, times cited, citations per year, impact for its field and year, cited by studies here, still being cited, senior author's track record, institution's track record, multi-centre collaboration and free to read, so those were left out: it earned 32 of the 56 points we could judge it on, which is 57 out of 100.
