Trem2R47H and reduced TREM2 expression both mimic human Alzheimer's disease signatures in mice
Tamar R Abel, Ravi S Pandey, Annat Haber, Dylan Garceau, Michael Sasner, Kevin P Kotredes, Gregory A Cary, Adrian Oblak et al.. bioRxiv : the preprint server for biology, 2025-12-19
Topic: Inflammation, microglia and astrocytes
Trem2*R47H mice had alternative splicing leading to reduced Trem2 expression; Trem2*R47HHSS mice expressed Trem2 at wild-type transcript and protein levels. Both models correlated with similar LOAD-associated signatures, and had similar effects on immune response, synapse, and vasculature biodomains. The Trem2*R47H model additionally affected extracellular matrix and myelination signatures. We demonstrated that Trem2*R47H and Trem2*R47HHSS mice are complementary models for the study of molecular contributions to LOAD pathology.

Key findings
- Trem2*R47H mice had alternative splicing leading to reduced Trem2 expression; Trem2*R47HHSS mice expressed Trem2 at wild-type transcript and protein levels.
- Both models correlated with similar LOAD-associated signatures, and had similar effects on immune response, synapse, and vasculature biodomains.
- The Trem2*R47H model additionally affected extracellular matrix and myelination signatures.
- We demonstrated that Trem2*R47H and Trem2*R47HHSS mice are complementary models for the study of molecular contributions to LOAD pathology.

How this study scored 16/100
Preprint — not yet peer-reviewed. It scored 22 on the same rubric, but non-peer-reviewed sources are capped at 16, so it shows as 16. Scored zero for study design, sample size, effect reported and randomised / blinded because the record does not state it. We have no data on times cited, citations per year, impact for its field and year, cited by studies here, still being cited, senior author's track record, institution's track record, multi-centre collaboration and free to read, so those were left out: it earned 13 of the 60 points we could judge it on, which is 22 out of 100.
