Knockdown of transactive response DNA-binding protein (TDP-43) downregulates histone deacetylase 6
Fiesel FC, Voigt A, Weber SS, Van den Haute C, Waldenmaier A, Görner K, Walter M, Anderson ML et al.. The EMBO journal, 2009-11-12
Topic: Brain proteins, microglia and lab models
TDP-43 is an RNA/DNA-binding protein implicated in transcriptional repression and mRNA processing. Inclusions of TDP-43 are hallmarks of frontotemporal dementia and amyotrophic lateral sclerosis. Besides aggregation of TDP-43, loss of nuclear localization is observed in disease.

Key findings
- Inclusions of TDP-43 are hallmarks of frontotemporal dementia and amyotrophic lateral sclerosis.
- Besides aggregation of TDP-43, loss of nuclear localization is observed in disease.
- To identify relevant targets of TDP-43, we performed expression profiling.
- Thereby, histone deacetylase 6 (HDAC6) downregulation was discovered on TDP-43 silencing and confirmed at the mRNA and protein level in human embryonic kidney HEK293E and neuronal SH-SY5Y cells.
Why it matters
This laboratory study in cells found that silencing TDP-43 lowers histone deacetylase 6, a finding relevant to frontotemporal dementia and ALS rather than Alzheimer's disease. It is early, cell-based work and reports nothing about treating, curing or preventing Alzheimer's disease.

How this study scored 45/100
Published in a peer-reviewed journal. Scored zero for study design, sample size, effect reported, studied in people and randomised / blinded because the record does not state it.
